Yes it's just a lens that focuses really closely. Technically to be a true macro lens it needs to be able to focus close enough to project the subject onto the sensor at the same size as real life (so the fly in the photo covers as much as if you put the actual fly into the sensor). This is called 1:1 magnification. Some specialist lenses can magnify further and let you photograph an insect's head to fill the full size of your sensor. You can turn a regular lens into a macro lens with extension tubes as it moves the sensor farther and lets you therefore focus closer. A macro lens that you buy will have better image quality though as it will have been designed differently in order to be optimized for focusing so closely. And focal length can be anything really. Typically around 100mm or so is common as it give more working distance from the subject, but there are some longer ones like 200mm and recently some wide angle macro lenses like from Laowa (which require the subject to practically touch the front element to reach 1:1 magnification).

It makes an enormous difference in my opinion and I use a diffused flash on basically all my 1:1 macro. Pros: lighting always looks good regardless of weather, location and time of day, you get base iso for max quality, you can shoot stopped down for depth of field, you can control light direction, freezes all movement from both the subject and camera shake. Cons: extra equipment to carry in your bag, in rare cases can have a distracting reflection on things like the carapace.

I like using natural light if I can for stuff that is not 1:1 though, like if I'm photographing a flower or shooting an insect in context. You can also shoot a mix of flash and natural light like getting a backlit subject in the setting sun and a lower powered flash mixed with it, though I've not explored that as much as I would like.

True I forgot to mention that. It's why I prefer natural light in images that are not true 1:1 macro as it shows context better with the scene. That said, I actually do quite like the black background from flash. It's nice and graphic and contrasty and can bring a nice mood. If you don't like it then with macro it's not difficult to put something behind for a splash of colour or texture, or dial in the settings to mix ambient and flash. Macro is one of the easiest forms of photography in the sense that so much is under direct control of the photographer.

For macro you don't need a fancy flash with TTL or anything as you'll be using it full manual. I use a cheap yongnuo flash I got for like $70. I think godox also makes some cheap ones. Unless you plan on using more advanced features for other types of photography it's better to buy a cheap flash in my opinion. Even if it breaks and you replace it you will be still spending far less.

Thanks. Increasing contrast is a tough issue when it comes to microscopes. There are a few solutions, I'm working on one right now.
In general one can do interference contrast, contrast staining, fluorescence microscopy or immuncytochemical marking. Or just make transgenic shit with GFP, but the plasmids are expensive and you can only use it ons something already in culture.
For now i'm just going to do DIC, where i take two polar filters, put one under the stage between the condensor and the light source and one above the stage where the head atatches to the arm. Rotate them at a 90 degree angle and only polarised light comes through. That increases contrast and makes membranes more pronounced.
Unfortunatly getting polar filters the right size and shape and quality is not cheap or easy.

When you're doing super macro, you'll have to use focus rack, ie focus by moving the camera as opposed of focusing with the lens. If you're around 1:1, you should be be able to get away with it even using automatic stackers.

How close do you have to get to the subjects? Particularly bugs. I'm wondering how easy it is to scare them off by needing to get really close with a macro. I'm used to shooting telephoto so it's not a consideration I have too often.
